Identification of Taxillus Chinensis Extract(Sang Ji Sheng Extract) Recorded in Chinese Pharmacopoeia
Take 5g of Taxillus Chinensis Extract(Sang Ji Sheng Extract) powder and raw Taxillus Chinensis as test sample and reference sample, add 60ml of methanol water (1:1), heat and reflux for 1 hour, filter while hot, concentrate the filtrate to about 20ml, add 10ml of water, and then add dilute sulfuric acid to about 0. 5ml, boil and reflux for 1 hour, then shake twice with ethyl acetate to extract 30ml each time. Combine the ethyl acetate solution and concentrate it to about 1ml as the test solution. Take another quercetin reference substance and add ethyl acetate to make it contain 0 in every 1ml. 5mg of solution is used as the reference solution. According to the thin layer chromatography method (Appendix VI B), extract 10μl of each of the above two solutions. Point on the same silica gel G thin layer plate prepared with 0.5% sodium hydroxide solution, use toluene (water saturated) – ethyl formate formic acid (5:4:1) as the developing agent, develop, remove, air dry, spray with 5% aluminum trichloride ethanol solution, and examine under ultraviolet light (365nm). In the chromatogram of the test sample, fluorescent spots of the same color appear at the corresponding positions as the reference sample chromatogram.
