Pharmacological action of Eucommia Extract Powder (Du Zhong)
1. The extract and decoction of tree bark have a long-lasting antihypertensive effect on animals. After intravenous injection of 5 milliliters of its extract (1-2 grams of raw medicine) into anesthetized dogs, it produced a significant antihypertensive effect that lasted for 2-3 hours and exhibited rapid tolerance. In three dogs with renal hypertension, a decoction of 5-8 g/kg was administered by gavage every day for a total of 4 weeks. The maximum reduction in systolic blood pressure was only 8-22 mmHg (4-10%), so the therapeutic effect was not satisfactory. The preparation and dosage form of Eucommia ulmoides have a certain impact on its antihypertensive effect.
The decoction has a stronger effect than the tincture, and the antihypertensive effect of fried Eucommia ulmoides is greater than that of raw Eucommia ulmoides. Eucommia ulmoides has a hypotensive effect on cats, but it lasts for a short time and the phenomenon of rapid tolerance is not significant. The antihypertensive effect on rabbits with cholesterol arteriosclerosis is more significant than on normal rabbits, but it can also produce rapid tolerance. As for the principle of blood pressure reduction, as the effective ingredients have not been determined yet, it belongs to preliminary properties. It has been proven that acute blood pressure reduction is not closely related to the vagus nerve and is not affected by atropine.
For isolated rabbit hearts, Eucommia ulmoides is initially excited at small doses and then slightly inhibited; There are also reports that its inhibitory effect on the heart is related to its pH (pH between 4-5), and after adjustment, it exhibits an excitatory effect on both rat and rabbit isolated hearts. Eucommia ulmoides has a direct dilation effect on normal rabbit ear blood vessels, but at the same concentration, it causes experimental cholesterol induced vasoconstriction in rabbit ear blood vessels. At low concentrations, it often exhibits dilation effect on normal rabbit coronary and renal vessels, while at high concentrations, it is the opposite. At low concentrations, it also exhibits contraction effect on coronary vessels in atherosclerotic rabbits. Because its blood pressure reduction is not directly related to the heart and blood vessels, it is inferred that it is central.
2. Diuretic effects: Various preparations of Eucommia ulmoides have diuretic effects on anesthetized dogs and there is no rapid tolerance phenomenon. It also has diuretic effects on normal rats and mice. Eucommia ulmoides contains 0.4% potassium, so it is inferred that diuresis may be related to potassium.
3. Other effects: Clinical use of Eucommia ulmoides extract can lower blood pressure in hypertensive patients and improve symptoms such as dizziness and insomnia. High doses (20-25 g/kg/day) of Eucommia ulmoides decoction administered to dogs by gavage can make them quiet, drowsy, and less susceptible to external stimuli. High doses also have an inhibitory effect on the central nervous system in mice. Eucommia ulmoides can inhibit the excitatory effect of the posterior pituitary gland on the isolated uterus of rats and rabbits, causing uterine relaxation. However, it has an excitatory effect on the isolated uterus of cats. Someone has reported that Eucommia ulmoides decoction has certain inhibitory effects on Mycobacterium tuberculosis in vitro. Its alcohol infusion seems to reduce the absorption of cholesterol in the intestine of rats.
